1785년 설립된 문리아카데미에서 출발했다. 당시 이 아카데미는 유서 깊은 스코틀랜드 대학의 민주주의에 기반한 교육이념을 따라 특정종교와 무관한 대학으로 설립되었다. 1800년 영국 성공회 계열의 뉴브런즈윅 대학(College of New brunswick)으로 인가를 받았다. 1859년 특정종교와 관련이 없는 종합대학으로 승격되어 뉴브런즈윅 대학교가 되었다. 

20세기 초 교과과정을 확대하고 공학관, 체육관 등 여러 건물을 지었다. 1960년대 학제를 개편했으며, 1964년 세인트존에 새 캠퍼스의 문을 열었다. 1973년 뉴브런즈윅 사범대학을 통합했다. 계속해서 단과대학을 늘리고 많은 건물을 신축하며 크게 발전했다. 2010년 가을 달루지 대학교, 뉴브런즈윅 주정부와 협력해 세인트존 캠퍼스에 의과대학을 설립했다. 이 대학은 북아메리카 지역에서 가장 오래된 공립대학 가운데 하나이자 캐나다에서 가장 오래된 공립대학 가운데 하나이다. 

현재 학예, 법학, 경영학, 교육학, 전산학, 임학, 환경관리, 과학, 공학, 운동과학, 간호학 분야 등의 14개 대학 및 대학원으로 구성되어 있다. 75개 이상의 분야에서 학사학위, 석사 및 박사학위를 수여한다. 특히 법학, 생의공학 분야의 명성이 높다. 인터넷 등 첨단통신매체를 이용해 원격교육을 실시하고 있다. 

2010년 기준으로 약 1만 2,000명의 학생들이 재학하고 있다. 전 세계 100개 이상의 나라에서 온 유학생들이 이곳에서 수학하고 있다. 교직원 수는 620여 명의 교원을 포함해 3,000명을 넘는다. 부속기관 및 시설로 도서관, 어학원, 체육관, 천문대, 대학신문사, 라디오방송국, 기숙사 등이 있다. 그리고 부설연구소로 캐나다사회정책연구소, 생의공학연구소, 행성우주과학연구소, 자기공명영상연구소, 만성질환연구소, 원자력에너지연구소, 캐나다강연구소, 전쟁·사회연구소, 정보기술연구소 등 60개 이상의 연구소가 있다. 

두 개의 주캠퍼스가 캐나다 뉴브런즈윅 주에 있다. 설립 당시 조성된 캠퍼스는 주도인 프레더릭턴에 있으며, 좀더 규모가 작은 캠퍼스는 주 최대의 도시인 세인트 존에 있다. 프레더릭턴 캠퍼스의 재학생 수는 9,000명이고, 세인트존 캠퍼스의 재학생 수는 약 3,000명이다. 그밖에 몽통과 배서스트에 보건 분야의 캠퍼스가 있으며, 카리브해 지역과 베이징에 사무소를 두고 있다. 

이 대학을 상징하는 건물 가운데 하나로 전쟁기념관이 있다. 이 건물은 원래 1924년 과학관으로 지어졌으며, 제1차 세계대전 당시 사망한 35명의 졸업생을 추모하는 뜻에서 전쟁기념관으로 이름을 바꾸었다. 현재 6만 4,000명 이상의 졸업생들이 전 세계 각지에서 활동하고 있다. 이 가운데 약 50%는 뉴브런즈윅 주에 거주한다. 유명한 졸업생으로 19세기 캐나다 시인인 블리스 카먼, 그래미상을 수상한 캐나다 가수 앤 머레이, 정치가인 앤디 스콧이 있다.

A first-class teaching and research institution, the University of New Brunswick (UNB) is one of the oldest public universities in North America. Located in Eastern Canada, UNB enrols nearly 13,500 undergraduate and graduate students. UNB’s two main campuses are located in Fredericton and Saint John. It also has satellite campuses and programs in other parts of Canada and the world.

UNB offers an international experience by attracting students from more than 100 countries. Students learn about different cultures through classes and special events on campus or participate in our study abroad programs to travel and study at the same time.

With 10 varsity sports teams competing in the CIS along with six varsity sports competing in CCAA, a large number of extra-curricular clubs and activities, and cultural offerings ranging from art galleries to resident musicians and writers, UNB offers a great student experience.
