간단히 ‘맥길’이라고 부른다. 영국 식민시대이던 1821년 몬트리올에서 활동하던 영국 스코틀랜드 출신의 모피상 제임스 맥길(James McGill)의 유언을 바탕으로 ‘맥길유니버시티칼리지(University of McGill College)’로 설립되었다. 1829년 기존의 몬트리올의과대학을 인수하여 맥길대학교 의과대학으로 개편하여 수업을 시작하였다. 1855년 유명한 지질학자인 38세의 젊은 윌리엄 도슨(William Dawson)이 총장으로 부임한 이후로 대학은 비약적인 발전을 이룩하였다. 1884년에 여성들의 입학을 허용하였다.
캐나다에서 역사가 가장 오래된 대학에 속하며, 연구 중심의 공립대학교이다. 2010년 기준 농업환경학·문리·치과·교육·의과·공과·경영·법과·음악·종교·과학의 11개 대학, 식이영영학·농업·사회사업·컴퓨터공학·간호·도시계획·환경·도서정보·통신과학·물리작업치료대학원의 10개 대학원이 있다. 그밖에 평생교육원을 운영한다. 강의는 영어로 진행된다.
몬트리올 번화가의 로열산(Mount Royal) 기슭에 위치한 주 캠퍼스와 주 캠퍼스에서 서쪽으로 30km 떨어진 생트안드벨뷔의 맥도널드 캠퍼스로 나누어진다. 맥도널드 캠퍼스는 맥도널드칼리지를 기부한 윌리엄 맥도널드 경에서 이름을 따왔다. 두 캠퍼스에는 레드패스자연사박물관(1880), 물리학동(1893), 레드패스도서관(1893), 화학동(1896) 같은 유서 깊은 건물들이 많다.
부설시설로 13개 도서관, 출판부, 수목원, 레드패스자연사박물관, 신경학연구소, 맥길건강센터, 몰슨스타디움 등이 있다. 5개의 별도 병원과 직접적으로 제휴하고 있으며, 몬트리올 소재의 여러 병원과 협력관계를 맺어왔다. 1980년대 말에 스튜어트생물학빌딩의 남쪽 부분 위쪽 2개층에 식물환경조절실험장치(phytotron)을 조성했졌다.
졸업생 또는 교수로 노벨상을 수상한 대표적인 사람은 어니스트 러더퍼드(1908, 화학), 윌러드 보일(2009, 물리학), 잭 쇼스택(2009, 의학)이 있고 캐나다 총리 윌프리드 로리에와 존 애벗, 라트비아공화국의 첫 여성 대통령 바이라 비케프레이베르가 같은 저명인사를 배출하였다.
